I'm not an expert on Mac's, but I believe you need a third-party program to do this, like Norton SystemWorks. Check this link to see if you really need to defrag:

i don't know that that is your problem normally it defrags it self
try clicking on your HD
click file drag to get info (cmd I)
then click index if your actually having problems
go to applications/Utilities/ find disk utility and find the button re-pare permissions
after doing these 2 steps it normally fixes all speed related issues

Yellow is right this program screws with your computer
ive installed it before on (OS 9) screwed the whole computer up
its speed disk doesn't change the location of recourse forks just the app. it self
on os X it worse messes with your main library it is like a viruses
also it sets up background tasks that slow down the computer
\and don't do any thing
Hmm... Have you checked Activity Monitor to see if there are any rogue processes eating up your CPU time?
